Three Entrances, Not One Park Road
The biggest thing to plan around is that Everglades National Park does not have one road running through it the way many parks do. The three entrances, Homestead, Shark Valley and Gulf Coast, are separate access points with no road inside the park connecting them, and each one is roughly an hour or more from the others by road. Pick your entrance based on what you want to do, not just which one is closest, since you cannot easily drive from one to another once you are inside.
Miami to Shark Valley
Shark Valley, on the Tamiami Trail (US-41), is the closest entrance to Miami: about 38 miles and 45 minutes west of downtown. It is built around a 15 mile paved loop road through sawgrass marsh, walked, biked or ridden on a tram, and it is the easiest entrance for a half day trip without much driving.
Miami to the Homestead Entrance and Flamingo

Which Entrance Should You Pick From Miami?
For a short visit with easy walking and biking, Shark Valley is the closest and simplest. For hiking trails, Flamingo and the classic park road, use the Homestead entrance and budget the full drive time south. Only head to the Gulf Coast entrance from Miami if boating or kayaking the Ten Thousand Islands is specifically what you are after, since the drive there and back eats most of a day on its own.
Planning Around the Drive
Because the three entrances do not connect inside the park, picking two in one day almost always means backtracking to a highway and driving around rather than through. If your trip includes both Miami and the west coast of Florida, it makes more sense to see Shark Valley or the Homestead entrance on the Miami side of the trip and save the Gulf Coast entrance for whenever you are already near Naples or Marco Island, rather than trying to fit all three entrances into a single Miami day trip.
